Claristep Filtration Assembly: A Novel Sample Preparation Method Suitable for the Analytics of Phthalates in Solid Environmental Samples
The filtration clean-up of particle laden environmental samples for phthalates analytics is a common step in sample preparation prior to GC-MS. Here, we demonstrate that Claristep???syringeless filtration units 0.45 μm and 0.2 μm are suitable for the filtration clean-up of phthalate samples. A mix of eight phthalates and one adipate was filtered at a concentration of 10 μg/mL with neither detectable adsorption nor releasing of these compounds from the filter unit. Furthermore, it was shown by GC-MS screening that no detectable compounds were extracted.
Impact of the Claristep? Filtration System on Recovery and Adsorption of Various Therapeutic Proteins at Low Sample Volumes
We tested the novel filter device Claristep? for the preparation of protein samples containing one of four different biotherapeutic molecules. In comparison to the standard protocols for sample preparation prior to analysis by either reversed-phase or SEC HPLC, the Claristep? filter did not exhibit a significant difference in protein concentration if the target molecules were present with at least 1.0 g L-1. A significantly reduced product concentration was observed in the Claristep? filtered samples compared to the standard protocol for a monoclonal antibody, if the concentrations were lower. The protein adsorption to the filter material was well described by a Langmuir adsorption isotherm. Additionally, the Claristep? filters facilitated a rapid sample preparation with minimal volume losses.

Ergonomic and Efficient Filtration for Food Analytics
In food analytics, clarification by membrane filtration is often an important step in sample preparation for processes like HPLC. Designed for parallel processing of up to eight samples, the Claristep filtration system was examined to determine its suitability for the filtration of food samples. Fourteen commercially available food samples – among them very particle-rich dispersions – were filtered at various levels of dilution with the syringeless Claristep? 0.45 μm and 0.2 μm pore size filters and the manual Claristep? base. Suitable dilutions were also tested with an established syringe filtration system, and the two techniques were compared with regard to ease and comfort of use. The results show which levels of dilution can be expected to result in successful filtration. In comparison to conventional filtration systems, testers consistently reported better ergonomic handling and significant time savings in sample preparation with Claristep.
